It acts by inhibiting the metabolism of folic acid in activated peripheral T cells. Specifically, methotrexate binds to and inhibits the enzyme dihydrofolate reductase, resulting in inhibition of purine nucleotide and thymidylate synthesis and, subsequently, inhibition of DNA and RNA syntheses.
